报告摘要:The hypothalamus-pituitary-gonadal (HPG) axis plays a critical role in the control of reproduction in teleost. In this axis, gonadotropin releasing hormone (GnRH) secreted from the hypothalamus initiates the cascade of events in stimulating luteinizing hormone (LH) and follicle stimulating hormone (FSH) synthesis and release from the gonadotrophs in the pituitary. LH and FSH act on the gonads to maintain their functional and structural integrity. Besides GnRH, several neuropeptides including kisspeptins, neurokinin b (NKB), gonadotropin-inhibitory hormone (GnIH) and spexin have been identified as positive or negative factors in the activation or suppression of the reproductive axis in teleost. However, because of the lack of a suitable genetic model, the in vivo functions of these genes on the fish reproductive axis are not fully elucidated. To address this issue, we have developed a TALENs system for efficient targeted gene knockout in zebrafish. Using our optimized platform, we have systematically mutated kiss1, kiss2, kiss1r, kiss2r, spexin, gnrh3, fsh鈒och, lh鈣, fshr, lhr in zebrafish. The obtained knockout zebrafish lines will provide insights into the neuroendocrine control of the reproductive axis in teleost.
